Wu, Gengshu, Peter Brouckaert, and Thomas Olivecrona. Rapid downregulation of adipose tissue lipoprotein lipase activity on food deprivation: evidence that TNFis involved. Am J Physiol Endocrinol Metab 286: E711–E717, 2004. First published December 23, 2003; 10.1152/ajpendo.00257.2003.—When food was removed from young rats in the early morning, adipose tissue tumor necrosis factor (TNF)activity increased 50% and lipoprotein lipase (LPL) activity decreased 70% in 6 h. There was a strong negative correlation between the TNFand LPL activities. Exogenous TNFfurther decreased LPL activity. Pentoxifylline, known to decrease production of TNF, had no effect on LPL activity in fed rats but almost abolished the rise of TNFand the decrease of LPL activity in rats deprived of food. The specific activity of LPL decreased from 0.92 mU/ng in fed rats to 0.35 and 0.24 mU/ng in rats deprived of food given saline or TNF, indicating a shift in the LPL molecules toward an inactive state. Lipopolysaccharide increased adipose tissue TNFand decreased LPL activity. Both of these effects were strongly impeded by pretreatment of the rats with pentoxifylline, or dexamethasone. Pretreatment of the rats with actinomycin D virtually abolished the response of LPL activity to food deprivation or exogenous TNF. We conclude that food deprivation, like lipopolysaccharide, signals via TNFto a gene whose product causes a rapid shift of newly synthesized LPL molecules toward an inactive form and thereby shuts down extraction of lipoprotein triglycerides by the adipose tissue.

Rapid downregulation of adipose tissue lipoprotein lipase activity on food deprivation: evidence that TNF-alpha is involved.

When food was removed from young rats in the early morning, adipose tissue tumor necrosis factor (TNF)-alpha activity increased 50% and lipoprotein lipase (LPL) activity decreased 70% in 6 h. There was a strong negative correlation between the TNF-alpha and LPL activities. Exogenous TNF-alpha further decreased LPL activity. Pentoxifylline, known to decrease production of TNF-alpha, had no effec...

Effect of tumor necrosis factor (TNF) on lipid metabolism in the diabetic rat. Evidence that inhibition of adipose tissue lipoprotein lipase activity is not required for TNF-induced hyperlipidemia.

Tumor necrosis factor (TNF) administration produces an increase in plasma triglycerides that may be due to inhibition of adipose lipoprotein lipase activity and/or a stimulation of hepatic lipogenesis. We now report that TNF administration to insulinopenic diabetic rats increases serum triglycerides (2 h, 2.4-fold; 17 h, 4.3-fold). Adipose tissue lipoprotein lipase activity was markedly decreas...

Lipoprotein lipase is produced, regulated, and functional in rat brain.

Lipoprotein lipase (LP lipase, triacylglycero-protein acylhydrolase EC 3.1.1.34) activity was found in four dissimilar brain regions (hypothalamus, cortex, cerebellum, and midbrain) of adult male rats. Progressive accumulation of LP lipase activity in cultured fetal rat hypothalamic cells was also observed, indicating de novo synthesis of the lipase. The brain LP lipase activity was serum-depen...

Two different mechanisms are involved in nutritional regulation of lipoprotein lipase in guinea-pig adipose tissue.

Lipoprotein lipase activity in adipose tissue responds rapidly to changes in the physiological state. To study what mechanisms are involved in the regulation, guinea pigs were fasted and the decrease in adipose-tissue lipoprotein lipase activity was compared with the decreases in mRNA and lipase synthesis. The mRNA pattern (three species) did not change. There was a close parallelism between th...

Lipoprotein lipase in the kidney: activity varies widely among animal species.

Much evidence points to a relationship among kidney disease, lipoprotein metabolism, and the enzyme lipoprotein lipase (LPL), but there is little information on LPL in the kidney. The range of LPL activity in the kidney in five species differed by >500-fold. The highest activity was in mink, followed by mice, Chinese hamsters, and rats, whereas the activity was low in guinea pigs. In contrast, ...
